Application notes for the separation of sarcosine, taurine and tetrahydrocannabinol metabolite using Cogent TYPE-C silica LC columns, are available from MicroSolv Technologies. Cogent TYPE-C silica LC phases offer unique opportunities for the separation of a wide range of sometimes difficult to separate compounds. They have the ability to retain polar solutes at high concentrations of organic solvent by aqueous normal-phase (ANP) and non-polar compounds under reversed-phase (RP) conditions.These columns use patented bonding technology to create a surface populated by silicon-hydride functional groups instead of silanols. The lack of surface silanols leads to fast equilibration times, excellent peak shape and extended column lifetimes for a wide range of analytes.
Cogent Phenyl Hydride for Analysis of Tetrahydrocannabinol Metabolite in Urine
9-Carboxy-11-nor-delta 9-tetrahydrocannabinol is the main metabolite of tetrahydrocannabinol (THC), formed in the body after consumption of cannabis. The compound stays in the body for a significant time, making it useful as a test compound for cannabis use. Following extraction of this cannabis metabolite from urine by SPE, it was successfully analysed by LC-MS using a Cogent Phenyl Hydride column, based on Type-C silica.
Separation of Sarcosine from Isobaric beta-Alanine
It is difficult to successfully separate sarcosine from isobaric beta-alanine under standard reversed-phase conditions without intensive sample preparation (i.e. derivatisation), as both compounds tend to elute at the solvent front. An alternative simple gradient LC-MS method, using a Cogent Diamond Hydride TYPE-C silica LC column shows the separation of these two compounds without the need for derivatisation. The developed method is sensitive, specific, quantitative and reproducible (%RSD = 0.1).
LC-MS Analysis of Taurine, a Polar Sulphonate Compound
Taurine is a non-essential sulphur-containing amino acid. It is used in food and pharmaceutical formulations, including many popular energy drinks. It is also found naturally in animal tissues and is a major constituent of bile. The high polarity of taurine creates a challenge for the reversed-phase HPLC of the underivatised compound. Cogent Diamond Hydride columns have been shown to provide excellent retention and peak shape under aqueous normal phase conditions for the LC-MS analysis of taurine.
